Astrios Cell Sorter

Despite having a number of available cell sorters in the core, the Astrios is by far the most sensitive at detecting small particles. The unique nature is the very low level of noise in the instrument, which is critical when we are looking for particles, with low fluorescence in the 50nm to 900nm range.

Unlike conventional analysers, when we are sorting we need to have the FSC trigger level as low as possible to ensure that we are not missing the smallest particles. This would normally allow a huge amount of noise to enter the system and flood the electronics.

  • FSC trigger off the FSC-1 at 0.005% shows 400eps as noise.
  • FSC triggered off the 405nm laser does not enhance anything on our system.
  • Running Megamix beads enable us to determine the noise and threshold rate based on size.
  • Megamix-Plus FSC is a mix of beads with the following diameters: 0.1 µm, 0.3 µm, 0.5 µm and 0.9 µm.
  • System pressure 60psi with a 0.5 pressure differential and an acquisition rate at 50,000 eps.
